Chloropyramine

Drug form and composition

Solution for injection. One ampoule of 2 ml contains 20 mg Chloropyramine Hydrochloride (10 mg/ml). Coated tablets. One coated tablet contains 25 mg Chloropyramine Hydrochloride. Ointment. One gram of ointment contains 1% 10 mg Chloropyramine Hydrochloride. Cream. One gram of cream contains 1% 10 mg Chloropyramine Hydrochloride.

Indications

As solution for injection Allergosan is used in acute states, associated with Quincke’s edema, allergic reactions after insect bites, drug and food allergy. In anaphylactic shock Allergosan is used as an element of the complex therapy. Tablets: Allergosan tablets can be used for the treatment of allergic rhinitis, conjunctivitis, hay fever, drug and food allergy, and allergic reactions after insect bites. Ointment or cream: Allergosan ointment or cream is used for the treatment of skin allergic reactions.

Dosage and administration

Parenteral administration: In cases of severe allergic reactions 1 to 2 ampoules can be injected intramuscularly or intravenously. Oral administration: In adults 1 tablet (25 mg) can be given 3 to 4 times daily (when necessary up to 150 mg); in over 5 years-old children Allergosan can be given in dose of 1 tablet 2 or 3 times daily. External application: The affected zone should be treated one or several times daily. During the application procedure ointment or cream should form a thin coating.

Contraindications

Parenteral or oral administration: Adenoma of the prostate, acute peptic ulcer, pyloric and duodenal stenosis, glaucoma, pregnancy, breast-feeding.

Special warnings and precautions

The medicine should not be used parenterally or orally together with alcohol, sedative drugs and hypnotics because of the potentiation of the effects. Allergosan should be used with caution in patients suffering from hyperthyroidism, cardiovascular diseases, and bronchial asthma. In children Allergosan can induce agitation and in many adult patients dizziness may be observed. Because of the pronounced sedative effect the preparation should be prescribed cautiously in drivers and people working with machines.

Drug interactions

Allergosan should not be used parenterally or orally concurrently with MAO inhibitors. Because of the anticholinergic activity of Allergosan concurrent administration with cholinomimetics is not advised. There is a possibility of increasing adverse cholinomimetic effects. General anesthetics, analgesic agents and psycholeptics potentiate the sedative effect of Allergosan.

Adverse reactions

After parenteral or oral administration Allergosan can induce: drowsiness, weakness, vertigo, tiredness, dryness in the mouth, constipation, and more rare reactions - visual disturbances and increase of intraocular pressure. In occasional cases, contact allergy and photosensibilization are possible after external application of the drug.

Pharmacological mechanisms

Allergosan is a representative of the classical antihistaminic agents - H1 antagonist. As a competitive reversible antagonist of histamine against the H1-receptors, Allergosan inhibits its effects. Allergosan inhibits the vasodilatation, increased vascular permeability and tissue edema. In addition to the antiallergic and sedative effects Allergosan also possesses a weak cholinolytic activity.

Supplied

10 or 100 ampoules of 2 ml (20 mg). 20 coated tablets of 25 mg. Tubes containing 18 g ointment (1%). Tubes containing 18 g cream (1%).

Expiry

Solution for injection and coated tablets - 5 years. Ointment and cream - 3 years.
